Our Science
We follow the biology to build better medicines.
Jade starts with validated biology and builds from there. Our programs are built on antibodies designed for potency, durability and tolerability, with the goal of translating scientific insight into clinical benefit for patients with autoimmune diseases.
How it’s designed:
JADE101 in IgAN
JADE101 is designed to target APRIL, a validated driver of pathogenic IgA production in IgA nephropathy.
IgA nephropathy is driven by long-lived plasma cells that produce an abnormal form of IgA. This initiates a cascade of immune activity that can lead to progressive kidney damage. APRIL plays a key role in supporting the survival and activity of these disease-driving plasma cells.
JADE101 was engineered to neutralize APRIL with ultra-high affinity at a novel epitope, inhibiting the signaling pathway that supports pathogenic plasma cells. By targeting the upstream driver of disease, JADE101 aims to reduce the production of disease-causing IgA while preserving normal immune function.
Our Targets
Targeting the biology
that drives disease
Jade is building a pipeline of novel monoclonal antibodies that target upstream immune pathways implicated in autoimmune disease. Each candidate is selected for clear biology rationale and designed for potency, durability, and tolerability, with a focus on outcomes that matter to patients.
JADE 101
anti-APRIL
APRIL is a cytokine that supports the survival of IgA-producing plasma cells. In IgA nephropathy (IgAN), the immune system produces an abnormal form of IgA that contributes to the formation of immune complexes in the kidneys, leading to inflammation and progressive damage. Because APRIL plays a central role in maintaining the plasma cells that drive this process, it represents a compelling therapeutic target in IgAN. Selective APRIL inhibition is intended to reduce pathogenic IgA and modify the underlying disease process without broadly depleting B cells.
JADE101 is a fully-human, ultra-high binding affinity, and extended half-life anti-APRIL monoclonal antibody in development for IgAN. JADE101 is designed to selectively neutralize APRIL with ultra-high affinity at a novel binding site, with the goal of inhibiting APRIL-mediated signaling and reducing pathogenic IgA production in patients with IgAN.
JADE 201
anti-BAFF-R
BAFF-R is a receptor expressed across B cell populations, including the autoreactive B cells that drive tissue damage in a wide range of autoimmune diseases. Signaling through BAFF-R supports B-cell survival signaling, maturation, and activation. In many autoimmune diseases, pathogenic B cells play a central role in driving chronic inflammation and tissue damage. BAFF-R inhibition offers a targeted approach to reducing these B-cell populations while also blocking the survival signals that help sustain them.
JADE201 is a half-life extended, afucosylated anti-BAFF-R monoclonal antibody designed to combine enhanced B-cell depletion with blockade of BAFF-R-mediated signaling. This dual mechanism is intended to support deeper and more durable B-cell suppression across a range of autoimmune diseases.
JADE 301
anti-IFN-β
IFN-β activity is believed to be a central driver of dermatomyositis (DM), a rare, chronic and debilitating autoimmune disease characterized by painful inflammatory skin disease and progressive muscle weakness.
JADE301 is a fully-human IgG1 monoclonal antibody with a YTE modification designed to selectively and potently neutralize IFN-β and inhibit downstream signaling implicated as a proximal driver in DM pathogenesis. Engineered with half-life extension technology, JADE301 is being developed with the goal of delivering a convenient, infrequent subcutaneous administration, with the potential to provide durable disease control and reduce treatment burden.
We advance what matters
The diseases we focus on are progressive, chronic, and underserved by existing therapies. Across autoimmune diseases, many patients continue to cycle through therapies that do not adequately control disease. This reality shapes how we develop medicines. Every scientific decision comes back to one question: how do we bring effective, well-tolerated therapies to patients with urgency and rigor?
Every program at Jade is guided with one goal: to advance therapies that have the potential to improve outcomes while fitting more easily into patients’ daily lives.
